RESUMO

The coffee fruit is preferably harvested at the cherry stage, with high moisture and metabolic activity, and must then undergo a drying process for better preservation of the bean and its sensory attributes. In this context, this study aimed to characterize the final quality of the Arara cultivar Arabica coffee processed using the wet method and subjected to six drying methods: three conducted at the agro-industrial establishment (fixed-bed dryer, rotary drum dryer, and combined drying) and three laboratory-scale methods (convective oven, cast-tape drying, and suspended terrace). Drying was carried out to reduce the coffee's moisture content from an initial value of 46.2% on a wet basis (w.b.) to a final average value of 11.35% (w.b.). The fruits of in natura demucilaged coffee and the processed dry coffee beans were characterized for moisture, ash content, nitrogen compounds, lipids, total titratable acidity, organic acids, sugars, and the instrumental color of the beans. The sensory profile of the Arabica coffee was evaluated by five coffee specialists using the methodology proposed by the Specialty Coffee Association (SCA), and all the coffees were classified as a specialty.

Aedes albopictus is a vector of several pathogens of significant public health concern. In this situation, gravid traps have become a common surveillance tool for Aedes spp., which commonly use hay infusions as an attractant. Diverse grass infusions have been assessed to enhance the attraction to this vector mosquito. However, these studies have focused on the oviposition effect, and the attraction potential to gravid Ae. albopictus females has not been evaluated yet. Here we report the attractiveness of infusions of 4 different botanical species (Cenchrus purpureus, Cyanodon dactylon, Megathyrus maximus, Pennisetum ciliare) as baits in sticky ovitraps and autocidal gravid ovitraps (AGOs) under laboratory, semifield, and field conditions. In the laboratory, Cynodon dactylon showed attractiveness, whereas in semifield conditions, both C. dactylon and Megathyrsus maximus were similarly attractive for gravid Ae. albopictus. None of the infusions conducted with AGOs were able to lure Ae. albopictus and other species of mosquitoes in a 14-wk field experiment. Our results demonstrate the feasibility of finding more attractive infusions for Ae. albopictus females to improve the efficacy of AGO traps, but further testing of infusions in AGOs in field settings is needed.

The bovine tick Rhipicephalus microplus, a primary ectoparasite of veterinary concern, contributes significantly to disease transmission and reduced cattle productivity, resulting in substantial economic losses. The overuse of chemical acaricides has led to the emergence of resistant strains, posing a considerable challenge to veterinary medicine. Consequently, the development of alternative parasite control methods is essential to ensure livestock quality and enhance food safety worldwide. Our study introduces an innovative approach to photodynamic inactivation (PDI) of the bovine tick, harnessing natural daylight for a potential field application. Reproductive parameters (female and egg mass, egg production index, and larval hatch) were evaluated in engorged female ticks under photodynamic action using the hematoporphyrin (HP) and tetra-cationic porphyrins free-base meso-tetra-ruthenated (4-pyridyl) (RuTPyP) and its zinc(II) complex (ZnRuTPyP) as photosensitizers (PS). The results showed that there was no significant difference between the groups treated with tetra­ruthenium porphyrins and the control group. However, HP exhibits a control percentage of 97.9% at a concentration of 2.5 µmol.L-1, aligning with the expected control rates achieved by conventional chemical acaricides. Photophysical and physicochemical parameters such as the number of singlet oxygen produced and lipophilicity were discussed for each PS and related to tick control percentages. Furthermore, the interaction between HP and chitin, an important macromolecule presents in the tick's cuticle, considered as the primary target tick structure during PDI was observed by the absorption and fluorescence emission spectroscopic techniques. Therefore, the results presented here extend the potential for controlling R. microplus through photodynamic inactivation while utilizing sunlight as a source of natural irradiation.

The design of rare-earth-doped upconversion/downshifting nanoparticles (NPs) for theoretical use in nanomedicine has garnered considerable interest. Previous research has emphasized luminescent nanothermometry and photothermal therapy, while three-dimensional (3D) near-infrared (NIR) luminescent tracers have received less attention. Our study introduces Nd3+-, Yb3+-, and Ho3+-doped NaYF4 core-shell luminescent NPs as potential multiparametric nanothermometers and NIR imaging tracers. Nd3+ sensitizes at 804 nm, while Yb3+ bridges to activators Ho3+. We evaluated the photoluminescence properties of Nd3+-, Yb3+-, and Ho3+-doped core and core-shell NPs synthesized via polyol-mediated and thermal decomposition methods. The NaYF4:NdYbHo(7/15/3%)@NaYF4:Nd(15%) core-shell NPs demonstrate competitive nanothermometry capabilities. Specifically, the polyol-synthesized sample exhibits a sensitivity of 0.27% K-1 at 313 K (40 °C), whereas the thermally decomposed synthesized sample shows a significantly higher sensitivity of 0.55% K-1 at 313 K (40 °C) in the near-infrared range. Control samples indicate back energy transfer processes from both Yb and Ho to Nd, while Yb to Ho energy transfer enhances Ho3+-driven upconversion transitions in green and red wavelengths, suggesting promise for photodynamic therapy. Fluorescence molecular tomography confirms 3D NIR fluorescence nanoparticle localization in a biological media after injection, highlighting the potential of core-shell NPs as NIR luminescent tracers. The strategy's clinical impact lies in photothermal treatment planning, leveraging core-shell NPs for (pre)clinical applications, and enabling the easy addition of new functionalities through distinct ion doping.

This work aims to assess the physicochemical characteristics and final sensory quality of Yellow Catuai IAC 62 Arabica coffee fermented with Saccharomyces cerevisiae. For such a purpose, a Composite Central Rotational Design (CCRD) was performed to investigate how fermentation time,temperature and pH conditions, moisture content and concentration of sugars and organic acids affect its sensory quality on two different roast levels in accordance with Specialty Coffee Association (SCA) protocols. It was found that fructose concentration decreased from 12 g/L to around 5 g/L during fermentation, regardless of temperature condition. Furthermore, longer fermentation times and higher temperatures have lowered sucrose and glucose concentrations from 4 to 2 g/L and 7 g/L to zero, respectively. Glycerol concentration was higher as time and temperature increased, and optimal conditions ranged at temperatures between 24 °C and 32 °C from 35 to 45 h of fermentation time. pH decreased as fermentation time elapsed, but there was a more significant reduction due to higher temperatures, starting at around pH 5 and, lower than 4 under extreme conditions. Contents of organic acids such as acetic, propionic, succinic, and lactic acids, were measured at the final stage of each fermentation process under studied conditions. It was observed that coffee samples achieved final scores ranging from 81 to 85 (SCA score), even in longer times and extreme temperature conditions, thus all samples have been classified as specialty coffees. This work described the initial step towards parameterizing fermentation processes, given that the response variables of temperature and fermentation time, were optimal and enhanced the sensory quality of coffee as beverage. Saccharomyces cerevisiae, a commercial product which has already been made available for producers, can ensure an increase in the sensory quality of coffee.

This work reports the structural characterization and photophysical properties of DyIII, TbIII, and EuIII coordination polymers with two phenoxo-triazole-based ligands [2,6-di(1H-1,2,4-triazole-1-yl-methyl)-4-R-phenoxo, LRTr (R = CH3; Cl)]. These ligands permitted us to obtain isostructural polymers, described as a 1D double chain, with LnIII being nona-coordinated. The energies of the ligand triplet (T1) states were estimated using low-temperature time-resolved emission spectra of YIII analogues. Compounds with LClTr present higher emission intensity than those with LMeTr. The emission of TbIII compounds was not affected by the different excitation wavelengths used and was emitted in the pure green region. In contrast, DyLMeTr emits in the blue-to-white region, while the luminescence of DyLClTr remains in the white region for all excitation wavelengths. On the other hand, EuIII compounds emit in the blue (ligand) or red region (EuIII) depending on the substituent of the phenoxo moiety and excitation wavelength. Theoretical calculations were employed to determine the excited states of the ligands by using time-dependent density functional theory. These calculations aided in modeling the intramolecular energy transfer and rationalizing the optical properties and demonstrated that the sensitization of the LnIII ions is driven via S1 → LnIII, a process that is less common as compared to T1 → LnIII.

In this work, a novel complex, [Dy(LPr)(NO3)2]·(H2O)·(NO3) (1), containing a highly distorted macrocyclic ligand (LPr) and weak axial anions (NO3-), was synthesized and characterized. Even though this coordination environment is not ideal for maximizing the magnetic anisotropy of a DyIII ion, a magneto-structural analysis reveals that the high distortion of the macrocycle promotes a disposition of the hard plane and easy axis opposite to the expected one. This results in a quite symmetrical environment which allows obtaining a field induced SMM behaviour. The magnetic relaxation properties of this complex were rationalized with the aid of ab initio multireference calculations. Moreover, 1 showed the characteristic emission bands of DyIII ion, indicating that the macrocyclic ligand acts as an efficient sensitizer in the energy transfer process to the emissive state of the DyIII ion. Due to the symmetric environment of 1, the Y/B intensity ratio (0.61) results in CIE coordinates (0.278; 0.314), close to those of the white light region. To gain further insight into the mechanism leading to the luminescence properties, ab initio calculations were performed to elucidate the key factors controlling the Y/B intensity ratio in this bifunctional complex.

This study aimed to redescribe two species of Ozolaimus, parasites of free-living green iguanas native to Marajó Island. The gastrointestinal system of four iguana specimens was evaluated for the presence of helminths. Altogether, 12,028 nematodes were found, with a prevalence of 100%, an infection range of 780 to 7,736 nematodes, an infection intensity of 3.007, and a mean abundance of 3,007. Light microscopy and scanning electron microscopy were used to determine the species of nematodes found. The cecum was the site of infection that had the highest parasitic load. Morphologically, the nematodes were compatible with the genus Ozolaimus Dujardin, 1844, with the species Ozolaimus megatyphlon (Rudolphi, 1819) Dujardin, 1845, and Ozolaimus cirratus Linstow, 1906. Scanning electron microscopy showed the presence of small structures (serrated in Ozolaimus cirratus and rounded in Ozolaimus megatyphlon) located below the esophageal leaves. We also evidenced the phasmids in both species; this is the first record of these structures in nematodes of the genus Ozolaimus. In addition, this work expands the records on the geographic distribution of these parasites.

Capillaria Zeder, 1800, parasitizes the organs and tissues of several hosts, including the domestic duck Cairina moschata (Linnaeus). This article describes a new species of Capillaria in domestic ducks identified based on morphological studies and molecular analyses of the ribosomal RNA gene. Thirty-eight specimens of C. moschata from the municipality of Soure, Marajó Island, Pará, Brazil. The organs of the birds' digestive tract were analyzed under a stereomicroscope to confirm the parasitic infection, after which the collected nematodes were identified by light microscopy, scanning electron microscopy, and molecular analysis. Capillariids parasitized the intestine and cecum of the examined birds. These parasites had three bacillary bands and a pair of elongated precloacal papillae on the tail. Phylogenetic analysis indicated that the new species formed a sister clade with Capillaria spinulosa (Linstow, 1890), as described in Indonesia and Japan. Based on morphological distinctions and molecular data, Capillaria cairina n. sp. can be considered a new parasite species of C. moschata in the Brazilian Amazon.

Caiman crocodilus is among the most abundant and widely distributed predators in the Neotropical region. These animals consume prey such as crustaceans, birds, small mammals, reptiles, amphibians, and fish, which can carry infective larval forms of nematodes. Brevimulticaecum has few studies on its morphology available, lacking detailed images. Therefore, the aim of this study was to redescribe Brevimulticaecum baylisi, stomach parasite of Caiman crocodilus, from subsistence hunting in the Yavari-Mirin River, Peruvian Amazon, using light and scanning electron microscopy. Four caimans were analyzed, and, macroscopically, all had ulcerative lesions in the stomach caused by this parasite. Histopathology showed an inflammatory infiltrate with a predominance of lymphocytes. Morphological characteristics of nematodes include the presence of three diamond-shaped lips wider than they are long, interlabia pyramidal, excretory pore located above the nerve ring, present intestinal cecum, ventriculus with five ventricular appendages, and long, winged spicules. These morphological characters, added to the number and distribution of the pre- and postcloacal papillae of the male specimens, allowed the identification of these parasites as B. baylisi. Scanning electron microscopy of these nematodes showed the presence of a dentigerous ridge on the inner surface of the lips in both sexes, while in males, the presence of a horseshoe-shaped median papilla was observed on the upper lip of the cloaca. Our research, therefore, adds these characteristics to the original description of B. baylisi, in addition to expanding the biogeographical distribution of this parasite.

The genus Baruscapillaria Moravec, 1982 has six valid species recorded in birds Phalacrocoracidae, namely Baruscapillaria appendiculata Freitas, 1933, B. spiculata Freitas, 1933, B. carbonis (Dubinin & Dubinina, 1940), B. jaenschi (Johnston & Mawson, 1945), B. phalacrocoraxi (Borgarenko, 1975) and B. rudolphii Moravec, Scholz and Nasincová, 1994. Helminthological tests carried out on cormorants of the species Phalacrocorax brasilianus (Gmelin), a migratory bird that occurs in the northeast of the State of Pará, Brazil, demonstrate B. appendiculata parasitizing the cloaca of these birds, through light microscopy, scanning electron microscopy and molecular biology. These studies allowed a redescription of males and females of this nematode in these hosts and in this geographical area through integrative taxonomy. The occurrence of lesions in the cloaca caused by this nematode parasite was registered using histological analysis. This is a new geographic report for this nematode.

For the first time in Brazil, Contracaecum australe is recorded parasitizing Phalacrocorax brasilianus (Aves, Suliformes, Phalacrocoracidae) from the Marine Extractive Reserve of Soure on Marajó Island, Brazilian Amazon. Its morphology revealed a body with a transversally striated cuticle, smooth or slightly cleft interlabia, lips with auricles, labial papillae, and conspicuous amphids. In males, the presence of the median papilla on the upper lip of the cloaca and spicules that reach almost half of the body of the parasite. These morphological characters, added to the number and distribution of the pre- and postcloacal papillae of the male specimens, and supported by the molecular phylogeny from the analysis of the ITS-1, 5.8S and ITS-2 genes, allowed the identification of these parasites.

RESUMO

We found serologic evidence of spotted fever group Rickettsia in humans and dogs and typhus group Rickettsia in dogs in Reynosa, Mexico. Our investigation revealed serologic samples reactive to spotted fever group Rickettsia in 5 community members, which highlights a potential rickettsial transmission scenario in this region.

Inducing immunogenic cell death (ICD) during cancer therapy is a major challenge that might significantly improve patient survival. The purpose of this study was to develop a theranostic nanocarrier, capable both of conveying a cytotoxic thermal dose when mediating photothermal therapy (PTT) after its intravenous delivery, and of consequently inducing ICD, improving survival. The nanocarrier consists of red blood cell membranes (RBCm) embedding the near-infrared dye IR-780 (IR) and camouflaging Mn-ferrite nanoparticles (RBCm-IR-Mn). The RBCm-IR-Mn nanocarriers were characterized by size, morphology, surface charge, magnetic, photophysical, and photothermal properties. Their photothermal conversion efficiency was found to be size- and concentration-dependent. Late apoptosis was observed as the cell death mechanism for PTT. Calreticulin and HMGB1 protein levels increased for in vitro PTT with temperature around 55 °C (ablative regime) but not for 44 °C (hyperthermia), suggesting ICD elicitation under ablation. RBCm-IR-Mn were then intravenously administered in sarcoma S180-bearing Swiss mice, and in vivo ablative PTT was performed five days later. Tumor volumes were monitored for the subsequent 120 days. RBCm-IR-Mn-mediated PTT promoted tumor regression in 11/12 animals, with an overall survival rate of 85% (11/13). Our results demonstrate that the RBCm-IR-Mn nanocarriers are great candidates for PTT-induced cancer immunotherapy.

Biomimetic nanoparticles hold great promise for photonic-mediated nanomedicine due to the association of the biological functionality of the membrane with the physical/chemical goals of organic/inorganic structures, but studies involving fluorescent biomimetic vesicles are still scarce. The purpose of this article is to determine how photothermal therapy (PTT) with theranostic IR-780-based nanoparticles depends on the dye content, cholesterol content, lipid bilayer phase and cell membrane type. The photophysical responses of synthetic liposomes, cell membrane vesicles and hybrid nanoparticles are compared. The samples were characterized by nanoparticle tracking analysis, photoluminescence, electron spin resonance, and photothermal- and heat-mediated drug release experiments, among other techniques. The photothermal conversion efficiency (PCE) was determined using Roper's method. All samples excited at 804 nm showed three fluorescence bands, two of them independent of the IR-780 content. Samples with a fluorescence band at around 850 nm showed photobleaching (PBL). Quenching was higher in cell membrane vesicles, while cholesterol inhibited quenching in synthetic liposomes with low dye content. PTT depended on the cell membrane and was more efficient for melanoma than erythrocyte vesicles. Synthetic liposomes containing cholesterol and a high amount of IR-780 presented superior performance in PTT experiments, with a 2.4-fold PCE increase in comparison with free IR-780, no PBL and the ability to heat-trigger doxorubicin release.

Considering the structural design of some of the scarce molecular-based Er-centred emitters in the literature, we explored the optical properties of three ErIII hexaazamacrocyclic complexes, namely Er-EDA (1), Er-OPDA(2) and Er-DAP(3). The macrocyclic ligands in these complexes differ in the lateral spacers, and are derived from 2,6-pyridine-dicarbaldehyde and ethylenediamine (EDA), ortho-phenylenediamine (OPDA) or 1,3-diaminopropane (DAP). Upon ligand-centred excitation, the bluish-green and green emissions of the ErIII ion were detected only for the complexes containing macrocycles with aliphatic spacers (1 and 3), which evidenced that these ligands can sensitize the ErIII luminescence. On the other hand, the ligand derived from the aromatic diamine (2) does not sensitize the ErIII luminescence. Energy transfer mechanisms, temperature sensing, CIE coordinates and CCT values were analyzed. Besides the excitation in the ligands, the erbium-centred excitation at 980 nm allowed the detection, in all cases, of bluish-green, green and red up-converted emissions, and also the downshifted NIR emission. The possible mechanisms involved in these transitions were described and analyzed according to the available data.

Objetivo: Determinar la incidencia de complicaciones de la técnica anestésica Spix en procedimientos odontológicos a pacientes atendidos en la clínica de la Universidad Andrés Bello (UNAB). Material y Métodos: Se analizó a 37 pacientes que fueron atendidos por alumnos de cuarto y quinto año de la clínica odontológica, a los cuales se le realizó la técnica anestésica Spix para realizar el procedimiento odontológico. Se consignó mediante la observación la presencia de formación de hematomas intraorales en el sitio de punción, rotura de la aguja, cantidad de tubos de solución anestésica inyectados, presencia de dolor a la inyección de solución anestésico y la presencia o no de parálisis facial. Mediante la recolección de datos y posterior encuesta a los participantes se consignó la presencia de trismus al día siguiente de la atención y parestesia persistente al día siguiente de la atención. Resultados: De 37 pacientes estudiados que recibieron la técnica anestésica Spix, 6 presentaron hematoma intraoral (16,2%), ninguno reportó rotura de la aguja, 1 presentó parálisis facial (2,7%), 1 presentó parestesia persistente al día siguiente (2,7%), 12 presentaron trismus posterior a la inyección (32,4%). El rango de dolor reportado fue entre 1 y 4 según la escala EVA. Conclusión: Hay una baja incidencia de las complicaciones asociadas a la técnica anestésica Spix en la clínica odontológica de la UNAB, siendo el trismus la complicación más frecuente. Se necesita un mayor número de muestra para entender mejor esta realidad.


Objective: To determine the incidence of complications of the Spix anesthetic technique in the dental procedures of patients attended at the Andrés Bello University dental clinic. Material and Methods: 37 patients who were cared for by fourth- and fifth-year students from the dental clinic of the Andrés Bello University were analyzed, who underwent the Spix anesthetic technique to perform the dental procedure. The presence of intraoral hematoma formation at the puncture site, needle breakage, number of injected anesthesia tubes, presence of pain upon injection of anesthetic and the presence or not of facial paralysis were recorded. Through data collection and subsequent survey of the participants, the presence of trismus was recorded the day after care. Results. Of the 37 cases of patients studied who received the Spix anesthetic technique, 6 had intraoral hematoma (16.2%), no needle break was reported, 1 had facial paralysis (2.7%), 1 had persistent paresthesia at the next day (2.7%), 12 presented trismus after the injection (32.4%), the pain range was between 1 and 4 according to the VAS scale. Conclusion. There is a low incidence of complications associated with the Spix anesthetic technique in the Andrés Bello University dental clinic, trismus being the most frequent (32.4%). A larger sample number should be needed to better understand this reality.

RESUMO

Two mononuclear DyIII complexes, [Dy(L1)(NCS)3] (Dy-EDA) and [Dy(L2)(NCS)3] (Dy-DAP), where Ln (n = 1-2) corresponds to a macrocyclic ligand derived from 2,6-pyridinedicarboxaldehyde and ethylenediamine (L1) and 1,3-diaminepropane (L2) were immobilized on functionalized silicon-based surfaces. This was achieved by the microcontact printing (µCP) technique, generating patterns on a functionalized surface via covalent bond formation through the auxiliary -NCS ligands present in the macrocyclic complex species. With this strategy, it was possible to control the position of the immobilized molecules on the surface. Water contact angle measurements, X-ray photoelectron spectroscopy (XPS), infrared reflection absorption spectra (IRRAS), and atomic force microscopy (AFM) confirmed that the surfaces were successfully functionalized. Furthermore, the optical properties in a broad temperature range were investigated for the as-prepared compounds. At room temperature, Dy-EDA was shown to emit in the deep blue region (Commission Internationald'Eclairage (CIE): (0.175, 0.128)), while Dy-DAP in the white region (CIE: (0.252, 0.312)). The different CIE values were due to the contribution of the strong emission of the ligand in the case of Dy-EDA. Besides, surface photoluminescence measurements showed that the immobilized complexes retained their bulk emissive properties.

RESUMO

The puffer fish Colomesus psittacus, is not commercialized on Marajó Island. They are captured as bycacth and discarded dead in the environment in artisanal fisheries that occur in the estuaries of northern Brazil. In this sense, the objective was to identify the parasites present in the gills and to evaluate the histopathological alterations caused by these nematodes of the genus Huffmanela. Fifty-five fish were analyzed, and thirty-five specimens showed the parasite in the gills. Morphological characteristics suggest that it is a new species of the genus Huffmanela, and the histopathological exams showed an edematous inflammation in the secondary lamella and the presence of eggs of this nematode, which is the first record of this parasite in C. psittacus in Brazil.
